====== It's day 1!! ====== **Contrast Agent Prep** - Gadovist, administered from 60mM diluted solution (diluted from 1M vials: sample dilution = 1.2mL 1M stock + 18.8mL saline) - administer at 5ul/g mouse, for total dose of 0.3umol/g - NEW lab standard (5Mar13): inject 5ul/g mouse + 60ul to accommodate 40ul dead volume + 20 ul flush Total: 5ul/g + 60ul **Catheter line prep** Components: - MTV-1 catheter with guidewire and 25G needle attached; filled with 3 units/mL Heparin - PE50 or PE20 tubing at customized length, according to mouse mass (see below for calculations) that has a 26G butterfly glued at one end and a 25G needle & syringe at other end; system contains Gadovist, though syringe & needle should be switched to contain saline to reduce dead volume - room line (bubble-free) containing saline; note it is ~12mL to flush entire line Set up is currently (18Aug2012): - MTV-1 catheter with 25G needle (included) and guidewire are filled with 3 units/mL Heparin - catheter inserted into vein, needle pulled back, tube glued onto tail, syringe pulled off & GUIDEWIRE pulled out - syringe re-attached to confirm flow into vein with heparinized saline - tube snipped to release syringe, 26G butterfly with PE tubing filled with Gadovist inserted, with saline-containing syringe at end - Everything secured for mouse transport to scanner - saline syringe removed, room line attached carefully! <code> (calculated as for PE-50):=(A3*5/1000)/(PI()*0.058^2/4) (calculated as for PE-20):=(B3*5/1000)/(PI()*0.038^2/4) PE50 tubing = 2.64ul/cm PE-50 PE-20 mouse length length weight Gd(cm) Gd(cm) (g) 18 34.1 150.2 19 36.0 158.5 20 37.8 166.9 21 39.7 175.2 22 41.6 183.6 23 43.5 191.9 24 45.4 200.2 25 47.3 208.6 26 49.2 216.9 27 51.1 225.3 28 53.0 233.6 29 54.9 242.0 30 56.8 250.3 </code> **Mouse Prep** - weigh - warm in chamber - mark tail for post-scan ID - catheterize - anesthetize: induce at 3%, reduce to 2% when asleep - transfer to coil bed, use lamp to keep warm - inject 0.5mL saline s.c. at scruff - administer eye gel - turn over, position s.c. tumour over / in coil - use mattress to prevent squishing?? - secure mouse & tumour using bellows & tape - secure catheter line with tape; syringe is ideally towards head end - insert and secure temperature probe (or do this at magnet) - turn off gas and transfer to magnet - **Coil Bed prep** - latex or nitrile glove piece taped over nose cone space securely, cut a small slit for insertion of mouse - use tube with no tooth bar to connect nose cone piece to isoflurane connector (black tube) - attach tubing for isoflurane vent - attach bellows to bed - make sure coil is ~approximately positioned according to nose cone, which can be secured with tape
